Joshua was overseer of the Berkley plantation during Bacon's Rebellion, was a member of the Governor's Council, and a member of the VA House of Burgesses. He made several trips to England trading tobacco, responsible for treaties with Indians from New York to VA, and mentioned numerous times as a close family friend of the Byrds of Westover (Colonel William Byrd II's diaries).

After building Fort Henry in 1646, he and three other Virginians, two servants and a guide explored what would become Tennessee and KY, being the first Englishmen to set foot there. After this, Joshua lived in Prince George Co., VA On 29 March 1715

Joshua Wynne and Robert Malone (husband of his niece Mary Wynne Malone) were paid bounty of 200 pounds to kill wolves. Joshua was also an Indian interpreter. He inherited his father Robert's plantation. He was a Justice in Charles City Co.,  VA, and also sheriff of Prince George Co.,  VA.

Major Joshua Wynne was shot and killed by Saponey Indians because one of Joshua's servants had killed one of the Indian's "great" men.  Upon trial of the Indian, they pleaded that the Wynne's were the aggressors and that they never rest without revenge. The Indians said that they and the Wynne's were then equal, each having lost a great man. To avoid more bloodshed the Indian was pardoned.

Joshua was an Indian interpreter and was said to be a fearless adventurer.
